Segundo os pesquisadores, o diferencial est\u00e1 na capacidade de superar mecanismos de resist\u00eancia desenvolvidos pelo microrganismo.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Embora o DAQ n\u00e3o seja uma mol\u00e9cula in\u00e9dita, j\u00e1 que sua atividade antimal\u00e1rica foi descrita ainda na d\u00e9cada de 1960, o grupo da Fiocruz coordenado pela pesquisadora Antoniana Krettli retomou os estudos utilizando abordagens mais recentes da qu\u00edmica e da biologia molecular.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cEssa mol\u00e9cula j\u00e1 tinha sido descrita como promissora, mas acabou sendo deixada de lado. O nosso grupo retomou esse estudo e mostrou um mecanismo \u00fanico de superar mecanismos de resist\u00eancia desenvolvidos pelo parasita, ao identificar uma caracter\u00edstica estrutural decisiva: a presen\u00e7a de uma liga\u00e7\u00e3o tripla na cadeia qu\u00edmica\u201d, explica Wilian Cortopassi, pesquisador colaborador da Fiocruz.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">O composto atua de forma semelhante \u00e0 cloroquina, interferindo em um processo essencial para a sobreviv\u00eancia do parasita. Durante a digest\u00e3o da hemoglobina humana, o microrganismo produz subst\u00e2ncias t\u00f3xicas que normalmente consegue neutralizar. O DAQ bloqueia esse mecanismo de defesa, levando \u00e0 morte do parasita.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Os estudos indicaram a\u00e7\u00e3o r\u00e1pida do composto nas fases iniciais da infec\u00e7\u00e3o e efic\u00e1cia tanto contra cepas sens\u00edveis quanto resistentes do&nbsp;Plasmodium falciparum. Os pesquisadores tamb\u00e9m identificaram resultados promissores contra o&nbsp;Plasmodium vivax, respons\u00e1vel pela maior parte dos casos de mal\u00e1ria registrados no Brasil.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Outro ponto destacado pelos pesquisadores \u00e9 o baixo custo potencial da mol\u00e9cula, fator considerado estrat\u00e9gico para pa\u00edses de baixa e m\u00e9dia renda, onde a mal\u00e1ria permanece end\u00eamica.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">As pesquisas contaram com colabora\u00e7\u00e3o de institui\u00e7\u00f5es como a University of California San Francisco (UCSF), a Universidade Federal de Alagoas (UFAL)&nbsp;e a Pontif\u00edcia Universidade Cat\u00f3lica do Rio de Janeiro (PUC-Rio). Novos estudos seguem em andamento em parceria com a Universidade Federal de S\u00e3o Paulo (Unifesp).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong>Apesar dos resultados considerados promissores, o desenvolvimento do DAQ como medicamento ainda depende de novas etapas, como testes de toxicidade, defini\u00e7\u00e3o de doses seguras e eficazes e desenvolvimento da formula\u00e7\u00e3o farmac\u00eautica adequada.<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Concedida em mar\u00e7o deste ano, a patente tem validade at\u00e9 5 de setembro de 2041. Para Antoniana Krettli, a estrutura da Fiocruz tamb\u00e9m pode acelerar futuras etapas de desenvolvimento do tratamento.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p class=\"wp-block-paragraph\">\u201cA institui\u00e7\u00e3o tem forte atua\u00e7\u00e3o na Amaz\u00f4nia, com diagn\u00f3stico e acompanhamento de pacientes, al\u00e9m de experi\u00eancia em testes cl\u00ednicos.
